Very simple, send a letter attaching a job description o he higher post. Obviously, the JD would contain the roles and responsibilities and also the minimum qualification required.

Promotion is management's preragative and it is not up to the employee to demand it. Call her in your office and verbally convey this as well as the fact that she does not possess requisite qualifications and meet other conditions.
