As per the Contract Labour (Regulation and Abolition) Act, 1970, the Principal Employer needs to obtain the Contract Registration Certificate if the laborers deployed by the contractor are more than 20. If the deployed laborers are less than 21, no Registration Certificate is required. Does this requirement still hold true?

The requirement was 20 or more contract workers deployed in an establishment. It was not less than 21; it was more than 20.

The number has since been increased to 50. So now in Karnataka (and more other states), you need a contract labor license if the contractor has more than 50 workers at a single establishment, and the establishment needs to register when it has 50 or more contract workers in total.
