Also the lady Hr. during conversation is very rude while I tried to oppose night shift by saying I m suffering with "migraine" she asked me that provide her in writing with doctor on his letterhead "not advisable for night shift" then they will think but not sure will retain.
Please advise me how do I refuse for night shift?

Dear Hrcnoida,
You say that you have medical problem and because of this, you would not like to do the night shift. When approached, HR person of your company has asked for medical certificate. But then what is wrong in her demand? How can HR take at its face value what employee says? Nevertheless, the method of telling this to you could be wrong. Probably she did not empathise with you and you felt that she was rude to you. Anyway, as HR says, provide the medical certificate which states that doing night duty could exacerbate your disease.
Next is about the night duty by other women employees. Will you be the first woman employee to do the night shift or few women employees are already doing the night shift? If few women are already working, then HR might have felt that if other women can, then why not you? Hence her assertiveness.
You have written that "company is bothering me for night shift". If the nature of your job demanded night shift then you could have been little circumspect while joining the company. While joining, were you not told that night shift was intrinsic part of your job?
The last sentence of your post is "Please advise me how do I refuse for night shift?" It appears that you are searching alibi to avoid the night shift. But then members of this forum take neutral stand and cannot be partisan to the person who has raised the query. Members' advice cannot be at the expense of the organisation to which the member belongs to!
Thanks,
